Research Context

SR-9009 was developed at The Scripps Research Institute and first described in 2012 as a synthetic ligand for the REV-ERB nuclear receptors, which regulate circadian gene expression and metabolic pathways. The original report characterized its effects on circadian behavior and on gene expression in liver, skeletal muscle, and adipose tissue in mice,^[1] and follow-up work examined REV-ERBα’s role in skeletal muscle mitochondrial biogenesis and oxidative capacity.^[2] It has also been used as a tool compound in cancer cell and oncogene-induced senescence models.^[3] Importantly for experimental design, later research found that SR-9009 affects cell proliferation and metabolism even in cells lacking both REV-ERB receptors, indicating off-target activity that complicates attributing its effects to REV-ERB alone.^[4]
